DNA-DNA hybridization was used to analyze the bacterial composition of three deep-sea sediment samples. Using a recA-based DNA probe we isolated a 16S rRNA gene from a related organism by colony hybridization of clones from the clones of the clone library from a deep-sea sediment. The clone selected (CHEF-DR-1416) showed an identity of 93% (cfu) when hybridized with the recA gene probe and of 96% (score) when hybridized with the 16S rRNA gene probe. The similarity of the 16S rRNA gene of CHEF-DR-1416 with known species was 83.1% with Thiomicrospira crunogena and 75.4% with Oligoflexia sp. All isolated clones showed a similarity of less than 78% with the clones previously isolated from deep-sea sediments. Hybridization of the clone with the recA gene probe resulted in a similarity of 61.4%.
